Rickettsia and Ehrichia from Ticks: field and laboratory investigation in Ganzhou City, Jiangxi Provence, China

Abstract:To understand the species of ticks and the pathogens they carry in Ganzhou City, Jiangxi Province, and to provide scientific basis for the prevention and control of tick-borne diseases. Ticks were collected in the field by flag cloth method and on the surface of animals, and the species were identified. The gltA gene and the 16s rRNA gene fragment of Rickettsia sp. and Ehrlichia sp. were amplified and sequenced by PCR, and the phylogenetic analysis was carried out. The species of 395 ticks collected from 4 counties of Ganzhou City, Jiangxi Province included 2 genus and 4 species, which were Haemaphysalislongicornis, H.concinna, H.cornigera, Rhipicephalusmicroplus respectively. Three hundred and ninety-five ticks were divided into 43 batches to detect Rickettsia sp. and Ehrlichia sp. A total of 18 ticks were positive. Phylogenetic analysis showed that 16 batches of ticks were positive in 22 batches of ticks in Longnan County, and LN1615 strains from H. cornigera were in the same branch as R.rhipicephali. The LN1620 strain from Rhipicephalusmicroplus was in the same branch as R.massiliae, while the other 12 strains came from H.cornigeras and 2 strains from Rhipicephalusmicroplus, which were in the same branch as R. japonica. The CY1602 strain in Chongyi County originated from H. concinna and R. raoultii was in the same branch. The YD1606 strain of Yudu County is Ehrlichia sp. from Haemaphysalislongicornis, which is in the same branch as Yonaguni206 (HQ697589), Yonaguni138 (HQ697588) and HLAE178 (GU075695) in 2009. The detection of Rhipicephalusmicroplus ticks in Anyuan County was negative. The ticks collected were identified in this paper. R.japonica, R. rhipicephali, R.massiliae, R.raoultii and Ehrlichia sp. were detected for the first time in Ganzhou City, Jiangxi Province. It provides the basis for the investigation of ticks in Jiangxi Province and the prevention and control of transmitted diseases.
