Research progress on polyethyleneimine as a nucleic acid vaccine adjuvant

Abstract:Vaccination is the most effective measure to prevent and control pathogen infection, and adjuvants are usually needed to ensure its effective immune responses. Polyethyleneimine (PEI), an organic cationic polymer, is traditionally used as the nucleic acid transfection reagent and the DNA vaccine delivery vehicle. In recent years, the adjuvanticity of PEI has been continuously documented. This review briefly summarizes the physicochemical characterization and general applications of PEI in distinct fields, and expounds the underlying immune activation mechanisms of PEI. We sum up the in vivo and clinical applications of PEI as a vaccine adjuvant, and discuss the challenges and future perspectives of PEI.
