Abstract:To establish a plaque detection method for reovirus, which laid the foundation for further study of the virus. Western blot and immunofluorescence were used to detect the infection of L929 cells and BSR cells with the Miyazaki virus strain of Nelson’s orthovirus, and the nucleic acid composition of the Miyazaki virus strain was verified by viral RNA horizontal gel electrophoresis. Establish a plaque detection method for reovirus and calculate the virus titer. The Miyazaki virus strain could successfully infect L929 cells. Horizontal gel electrophoresis verified that the nucleic acid composition of the virus strain was correct. L929 cells infected by the virus strain had plaques. The virus titer was calculated to be 2.2×108 PFU/mL. The detection method of reovirus plaque was successfully established. This method can form clear and visible plaques, which can quantitatively determine the virus titer stably and provide help for the further study of reovirus.
