Abstract:This study analyzed the epidemiological characteristics of H3N2 subtype influenza virus and the molecular characteristics of the neuraminidase (NA) gene from 2017 to 2019 in Guizhou Province, to provide a scientific basis for the prevention and control of H3N2 subtype influenza virus. Pharyngeal swabs of influenza-like cases (ILI) from 2017 to 2019 were collected from 13 national influenza surveillance sentinel hospitals in Guizhou Province and detected by RT-PCR. Statistical analysis was conducted on the basis of the monitoring cycle of natural year. The H3N2 subtype positive nucleic acid samples were isolated from dog kidney germ cells (MDCK), and the culture medium was collected to extract the viral RNA. After RT-PCR amplification, the products were purified for sequencing. The homology, genetic evolution and drug resistance sites were analyzed with MAGE6.0 software and ClustalW2 software. From 2017 to 2019, 15 553, 15 164 and 15 348 ILI throat swab samples were collected in Guizhou Province, all of which were nucleic acid tested. The positivity rates of the H3N2 subtype were 5.12% (797/15 553), 0.80% (122/15 164) and 7.67% (1 177/15 348), respectively. The peak period was mainly January-March and October-December, in agreement with the findings in the entire country. From 2017 to 2019, the nucleotide homology of the H3N2 subtype NA gene was 97.5%-100% and was mainly divided into two branches in the evolutionary tree. No mutations occurred in the key sites of the NA gene in all strains. In Guizhou province, the H3N2 subtype influenza virus manifests as a yearly periodic epidemic, mostly in winter and spring. To better prevent and control influenza and understand the epidemic trends in a timely manner, monitoring and rapid analysis of the genetic variation of the virus must be intensified.
