Abstract:The purpose of this study is to identify,clone and express the novel proteasome gene of Clonorchis sinensis and to predict the structure and function of the genes with bioinformatics methods in order to provide the foundation for functional study.From a cDNA library of Clonorchis sinensis,the novel proteasome gene was identified by screening the whole length cDNA with large-scale sequencing.The bioinformatics softs include the ORF finder,BLAST,Conserved Domains and ExPASy to analysis the structure,function,homology,phylogeny of the gene and putative proteins.The coding region of PSMA2 was amplified by PCR and then cloned into pGEX-4T-1.After induction with IPTG,the fusion protein was expressed and identified by SDS-PAGE.The result showed that a novel proteasome α2 gene of Clonorchis sinensis was identified(GenBank Accession NO.:EU223819).The 833bp CsPSMA2 gene encoded a putative protein of 235 amino acids with a predicted molecular mass of 26.0 kDa.Between the 5-233aa there was a whole proteasomeα 2 domain.Phylogenetic analysis indicated that the CsPSMA2 was close to the PSMA2 gene of Schistosoma japonicum.It is evident that a recombinant plasmid was constructed for expressing the GST fusion protein,and a novel CsPSMA2 gene was identified.Its key domain and structure was described.
