Abstract:There were three epidemic peaks of human infection with avian influenza A (H7N9) in mainland China in winter and spring since 2013. In this study, we established the assay of amplification and sequencing to obtain the complete genome of avian influenza virus A (H7N9). After the RNA from chicken embryo-eggs isolated strains or throat swab specimen was extracted, 22 fragments of genome were amplified by RT-PCR and sequenced with a set of self-designed primers. The complete genome sequence was obtained by sequence assembly and analyzed via bioinformatics. Each of the 8 genes of virus was closely related to that of H7N9 subtype viruses currently epidemic in mainland China. In conclusion, the assay could be used for amplifying and sequencing the complete genome of avian influenza H7N9 subtype virus effectively, which could lay the foundation for the further analysis of gene characteristics.
