Effects of gene expression profiles of A549 cells induced by Toxoplasma effector ROP16Ⅰ by using bioinformatics analysis

Abstract:To explore the differential genes in the progress of Toxoplasma gondii (T. gondii) type Ι and providing a basis for revealing pathogenesis of T. gondii type Ι, we analyzed the results of transcriptional profile of genes in the ROP16Ι-transfected A549 human lung adenocarcinoma cell line using bioinformatics methods. In this study, we used bioinformatics to identify genes that were differentially expressed. Then, ToppGene and STRING were performed on screened differentially expressed genes, and the Gene Ontology(GO) functional classification and KEGG analysis were conducted. 483 differential expressed genes were analyzed,among which 252 genes were up-regulated and 231 were down-regulated. Besides, a total of 14 candidate genes from the DEGs were screened using the ToppGene Suite of which CCL8,CXCL11,PIAS1and EIF2AK2 are related to T. gondii type Ι. GO analysis and KEGG analysis indicated that the DEGs were closely associated with interleukin IL-10, IL-4, IL-13 signaling pathway, cytokine receptor interaction pathway and Toll-like receptor signaling pathway. In addition, these 14 candidate genes also happened to be at the core position of the network in protein-protein interactions. Taken together, pathogenesis of T. gondii type Ι may be closely related with CCL8,CXCL11,PIAS1 and EIF2AK2 genes, but the exact mechanism is little elucidated, which needs to be further explored.
