HSP70及NF-κB在金葡菌致人巨噬细胞系U937细胞凋亡中的作用

Roles of HSP70 and NF-κB in Staphylococcus aureus-induced apoptosis of human monocytic U937 cells
摘要 目的探讨热休克蛋白70(HSP70)及核因子-κB(NF-κB)在金黄色葡萄球菌(简称金葡菌)诱导人巨噬细胞系U937细胞凋亡过程中的作用。方法当细胞∶细菌为1∶20时分别培养0min,15min,30min,60min和90min时,用台盼蓝染色检测金葡菌感染不同时间后U937细胞的存活率;用Western blotting方法检测热休克蛋白(HSP70)及NF-κB蛋白的表达。结果U937细胞的存活率呈时间依赖性降低;HSP70的表达随着金葡菌感染时间的延长逐渐升高,而NF-κB的表达随着金葡菌感染时间的延长逐渐降低。结论金葡菌诱导U937细胞凋亡过程可能与HSP70和NF-κB表达的改变有关。

Abstract :To investigate the relationship between the induced apoptosis and expression of heat stress protein 70kDa (HSP70) and nuclear factor kappa B (NF-κB) in human monocytic U937 cells by Staphylococcus aureus(S.aureus),U937 cells were used in all experiments,and they were infected with S. aureus for different lengths of time (0 min,15 min,30 min,60 min and 90 min respectively) at a ratio of U937/S. aureus of 1∶20. The necrosis rates of U937 cells were examined by trypan blue staining and the expression of HSP70 and NF-κB was detected by Western blotting. The results showed that the percentage of the necrotic cells was raised with the increasing of time. and S. aureus could induce the activation of HSP70 in a time-dependent manner and also down-regulate the expression of NF-κB. It is concluded that U937 apoptosis induced by S. aureus may be related to the increase of HSP70 and decrease of NF-κB.
