Abstract:The aim of this study was to investigate the role of Xlinked inhibitor of apoptosis protein (XIAP) and cIAP1/2 in the apoptosis of human monocytic U937 cells induced by Staphylococcus aureus (S. aureus). U937 cells were treated with S. aureus at different time points, and Western blot were performed to detect the expressions of XIAP and cIAP1/2. The U937 cells were pretreated with Embelin (an inhibitor for XIAP), and the apoptosis rates of U937 cells were detected by Annexin V FITC/PI assay. The results showed that the downregulation of XIAP and cIAP1/2 by S. aureus were statistically significant and inhibition of XIAP with Embelin caused further increase in apoptosis of U937 cells. It is concluded that XIAP and cIAP1/2 participated in the apoptosis of U937 cells. Embelin increases U937 cells apoptosis induced by S. aureus via inhibiting the expression of XIAP.
