Abstract:Abstract Objective: To reconstruct the three-dimensional image of Demodex folliculorum using confocal Laser scanning microscope (CLSM). Methods: The specimens were stained initially with fluorescein isothiocyanate (FITC) and then propidine iodide (PI) for a secondary fluorescent staining. A three-dimension image was reconstructed to observe and evaluate the morphological characteristics of the Demoded folliculorum. Results: The Specimens were well combined with fluorescein isothiocyanate and propidine iodidel. By CLSM, the real and complete morphology of the mited were exposed by the high definition three-dimensional pictures obtained with the continuous optical slices of the specimens. Conclusion: Both FITC and PI have a good fluorescence staining effect on the mite; meanwhile, the three-dimensional image of the mite obtained using CLSM can show a more precise ultrastructure, and the three-dimensional reconstruction technology has significant value for extensive application in small organisms.
