Abstract:ABSTRACT:To investigate the expressions of osteopontin and interleukin-10 and their correlation in Gerbil hepatic alveolar hydatid tissue, 60 gerbils were infected with 20% E.multilocularis suspension (0.1 mL per gerbil) through abdominal opening injection in liver. They were randomly divided into model group (No intervention, 40 gerbils) and experimental group (Anti-OPN antibody interference, 20 gerbils). Gerbils were sacrificed at 100 days post-infection for observing the growth and metastasis of hepatic Echinococcus cyst. The expressions of OPN and IL-10 in hepatic Echinococcus cyst and metastasis tissue were measured by immunohistochemistry staining (SP method). Results showed that all gerbils infected with hepatic alveolar hydatid. Expression of OPN and IL-10 were found at different degree in Echinococcus cysts. In model group, the positive expression rate of OPN and IL-10 in Echinococcus cysts were 72.5% (29/40) and 65% (26/40). Both OPN and IL-10 were mainly distributed in the germinal layer. The 65% (26/40) of them occurred thoracic lymph node metastases. The OPN and IL-10 expression rate in hepatic Echinococcus cysts with thoracic lymph node metastases were 84.6% (22/26) and 76.9% (20/26), which were significantly higher than that of hepatic Echinococcus cysts without thoracic lymph node metastases (50%, 42.9%) (P<0.05). In experimental group, the positive expression rates of OPN and IL-10 in Echinococcus cysts were 40% (8/20) and 35% (7/20). The expression of OPN was significantly correlated with the expression of IL-10 (r=0.605, P=0.000). In conclusion, OPN might significantly relate to the metastasis of hepatic Echinococcus cysts. OPN and IL-10 might play synergetic roles in hepatic alveolar hydatid tissue.
