Abstract:ABSTRACT: To investigate the mechanism of CVB3 damage on Golgi apparatus and discuss the pathogenic mechanism of CVB3 infection.After infecting Hela cells with CVB3 of MOI 5,we observed the variation of Golgi ribbon by detecting the locations of viral protein VP1 cellular proteins GM130 and GRASP65 in Hela cells at different post-infection times with the double labeled immunofluorescence technology.The total cellular proteins were collected for Bradford quantitative analysis at different post-infection times ,then the level of the viral protein VP1 cellular proteins GM130 and GRASP65 expression in Hela cells were analyzed by Western blot.The immunofluorescence of GM130 and GRASP65 were ribbon-like and the structure of Golgi apparatus was complete in the normal Hela cells; but the immunofluorescence of GM130 became dispersed and the structure of Golgi apparatus was damaged in the infected cells from the time of 3h post-infection. The level of cellular proteins GM130 and GRASP65 expression decreased gradually from the time of post-infection, the high level of the viral protein VP1 expression had lasted since Hela cells were infected with CVB3 for 6 h. The level of GM130 and GRASP65 expressions were down-regulated after CVB3 infection, which induced diffusion in the immunofluorescence of GM130 and GRASP65, and disruption in the Golgi ribbon. It might be a possible pathogenic mechanism inducing the host cells Golgi apparatus damage after the infection of CVB3.
