Abstract:Abstract:In order to further study the relationship between the cell division and the disaggregation of yeast prion [PSI+] with the treatment of guanidine hydrochloride, we investigated effect of hydroxylurea on guanidine hydrochloride curing yeast prion [PSI+] with a recombinant Sup35-GFP strain using the techniques of phenotype analysis and semi-denaturing agarose gelelectrophoresis (SDD-AGE) combined with western blot. Our results showed that guanidine hydrochloride was unable to cure yeast prion [PSI+] when cell division was suppressed by hydroxylurea. Further research showed that hydroxylurea could significantly decrease the ability of guanidine hydrochloride to disaggregate yeast prion [PSI+]. These results suggested that cell division is essential for the curing of yeast prion [PSI+] by guanidine hydrochloride.
