Abstract:Objective: In order to provide a base for the preparation of oral gene vaccines, the fusion expression vector of G gene and N gene of Rabies virus was constructed and was transformed into Saccharomyces cerevisiae expression system. Method: The plasmid pVax-G was used as a template for amplifying G gene and N gene of Rabies virus. Then these two genes linked together and were ligated with Saccharomyces cerevisiae expression vector. After identified by sequencing, this fusion expression vector was transformed into Saccharomyces cerevisiae expression strain INVScI to induce the expression of pVax-G/N fusion protein. Results: The sequencing result of fusion gene pVax-G/N was absolutely accorded with our anticipation. And the SDS-PAGE and Western-blotting analyses indicated that the induction of expression of fusion protein by galactose was successed. Conclusion: Successfully constructed and expressed the fusion expression vector pYes2-pVax-G/N, and this study provided a base for the preparation of oral Rabies virus vaccines which would use Saccharomyces cerevisiae as a transport vector.
