Abstract:Objective To investigate natural epidemic focus of Orientia tsutsugamushi in Bole area of Xinjiang, china. Methods DNAs were extracted from spleens of the captured mice and the blood of the worker and the Chigger mite were seized from the rodent body surface. Nested-polymerase chain reaction (nPCR) technique was used to detect the Ot-Sta 56 gene. Some positive samples were sequenced and analyzed by NCBI and analyzed in BLAST. Results We detected the positive from the rodents、worker and the Chigger mite. Orientia tsutsugamushi were isolated from the rodents spleen and trombidiid. The prevalence of Ot were 5.55% and 8.3% respective. The Sta56 gene nucleotide sequence homology to Karp strain was 99%. Conclusion The natural epidemic focus of Ot could exist in plain and wetland in Bole area. The host could be M.musculus、C.migratorius、R.opimus. The medium could be L.bolei .
