Abstract:The purpose of this study was to observe the clarithromycin resistance rate of Helicobacter pylori and to analyze gene mutations related with clarithromycin resistance in Guiyang area.Clarithromycin susceptibility of Hp clinical isolates were detected by agar dilution method.The 23S rRNA domain V gene fragment of Hp strains were amplified and sequenced. These strains included 10 clarithromycin resistant clinical isolates ,4 clarithromycin susceptible clinical isolates, Hp SS1 and Hp NCTC 11637.The sequences of these strains were analyzed by CLC Free Workbench 4.5.The results showed that the clarithromycin resistance rate of Hp in Guiyang was 30.9%. Five types of mutation were found in 10 clinical isolates.These mutations included:T2183C(10/10); T2245C(9/10);A2144G(6/10);C2196T(1/10);A2224G(1/10).But except A2144G, other mutations were also found in 4 clarithromycin susceptible clinical isolates.The results suggested that the clarithromycin resistance rate of Hp in Guiyang was high, A2144G was the main characteristic of gene mutation related with clarithromycin resistance in Guiyang.
