Immunocompetence of Helicobacter pylori ureI DNA vaccine in mice

Abstract:The aim of this study is to investigate the humoral and cellular immune response in C57BL/6 mice induced by the Helicobacter pylori urea channel protein gene (ureI) DNA vaccine. C57BL/6 mice were immunized with pcDNA3.1(+)/ureI or pcDNA3.1(+) or PBS buffer intramuscularly at 2-week interval for four times. ELISA was employed to quantitatively detect the levels of specific IgG antibody in the sera of mice and the contents of IL-4 and IFN-γ in mice spleen lymphocyte culture supernatant. The proliferation of mice spleen cells was evaluated by MTT assay, and the expression of ureI in musculature was analyzed by PCR and immunofluorescence histochemistry. Results demonstrated that the pcDNA3.1(+)/ureI DNA vaccine induced specific IgG antibody production in mice. After stimulation by UreI recombination protein, the stimulation index and the contents of IFN-γ and IL-4 in cultural supernatant of spleen lymphocytes from mice immunized with pcDNA3.1(+)/ureI increased significantly. The ureI gene could be expressed in mouse muscular tissue as determined by PCR and immunofluorescence. These results suggested that pcDNA3.1(+)/ureI DNA vaccine administered intramuscularly could induce strong humoral and cellular immune response in C57BL/6 mice, which lay a foundation for further study of the protective immunity against Helicobacter pylori of this DNA vaccine.
