Mosquito-borne arbovirus investigation in Yunfu Port, China

Abstract:Mosquito samples were collected at Yunfu Port in China and detected for representative mosquito-borne arboviruses during May to November of 2011. Representative mosquito-borne arboviruses, including dengue, Japanese encephalitis (JE), chikungunya, West Nile, and yellow fever viruses, were detected by multiple real-time fluorescence PCR assay. A further study of PCR amplification and sequence analysis to the positive samples were performed. A total of 39 254 individual mosquitoes were collected from 460 mosquito pools at Yunfu Port. All the virus including dengue, chikungunya, West Nile, and yellow fever viruses showed negative results except for 14 positive results with JE virus (JEV) nucleic acid detected by real-time PCR assay. The genotype of JEV was confirmed as genotype Ⅰ by PCR amplification and sequence analysis of E gene. The genotype Ⅰ JEV carried by mosquito was found for the first time in Guangdong. The minimum infection rate (MIR) of JEV in Culex tritaeniorhynchus was 0.937‰, which was slightly higher than that in Anopheles sinensis at 0.385‰ at Yunfu Port. Although the difference in MIR was not statistically significant, the outbreak and epidemic risk of mosquito-borne arboviral disease still exists. The primary investigation of mosquito-borne arbovirus at Yunfu Port would be useful to offer an effective method for future mosquito-borne arbovirus surveillance and detection.
