Abstract:The objective of this study is to obtain polyclonal antibody against the p38 protein of Echinococcus granulosus (Eg) in order to provide antibody for further study on the function of Egp38 protein. The full sequence of Egp38 gene was successfully cloned into the expression vector pET28a, and proved by restriction endonuclease digesting and sequencing. Then, the expression plasmid was transformed into E. coli BL21 and induced by 0.2 mmol/L IPTG, and then a 46 kDa Egp38 protein was expressed. The rabbit was vaccinated with the Egp38 protein, which was purified by affinity chromatography. ELISA and Western blotting showed that the titer of the anti-serum was above 2.56×105, and the anti-serum could specifically bind with Egp38 purified protein and native Egp38 in the germinal layer and the protoscolex of Echinococcus granulosus. The results indicated that the polyclonal antibody against the Egp38 was successfully prepared, which laid a foundation for further study on the function of Egp38 participate in the interplay between Echinococcus granulosus and intermediate host and the potential value as drug target.
