Ultrastructure of the integument and haemocoele in [WTHX]Dermatophagoides farinae (Hughes) (Acariformes: Pyroglyphidae)

Abstract:In this study, we observed the ultrastructure of the integument and haemocoele for adult Dermatophagoides farinae with a transmission electron microscope. Twenty mites were first fixed by using 5% glutaraldehyde and postfixed by using 1% osmium tetroxide. The TEM observation was conducted after dehydration, embedding, ultrathin section, and positive staining were finished. Results showed that the curticle of D. farinae consisted of epicuticle and procuticle, in which the epicuticle is further divided into cuticulin layer, wax layer and cement layer. The procuticle is composed of endocuticle and exocuticle, which was laminated obviously and there were many pore cannals and lamellae in it. The haemocoele of Dermatophagoides farinae was surrounded by the integument, various tissues and organs are immersed in it. There were a large number of mitochondria, vacuoles, glycogen granules, and a few bacterias and phagocytes in the hemolymph. The ultrastructural observations of the integument and the haemocoele provided a more comprehensive experimental basis for the morphology research of D. farinae.
