Serogroup identification and analysis of Leptospira isolates using serogroup specific PCR in Guizhou Province, China

Abstract:In order to understand the epidemic serogroup of pathogenic Leptospira and provide technical methods and scientific basis for the control and prevention of leptospirosis, the serogroup of Leptospira isolates collected in Guizhou Province were identified and analyzed by using O-antigen gene based serogroup-specific PCR. A total of 58 strains of leptospires collected from Guizhou Province in recent years were detected with pathogenic leptospire specific PCR (G1/G2-PCR) and further identified using O-antigen gene based PCR (O-PCR) which is specific for the serogroups of pathogenic Leptospira. Results were then confirmed with conventional microscope agglutination test (MAT). G1/G2-PCR detection results showed that all of the 58 isolates were pathogenic leptospires. O-PCR identified the 58 isolates as Leptospira serogroup icterohaemorrhagiae, which was consistent with the results of conventional MAT. Results of this study indicate that O-PCR can be used as a reliable tool for Leptospira serogroup identification in Guizhou Province, and the results of O-PCR indicated that Leptospira serogroup icterohaemorrhagiae was the epidemic serogroup of Leptospira in Guizhou Province in recent years.
