Abstract:To investigate the roles of colicin receptor CirA played in Escherichia coli involved antibiotic-resistance. λ-Red homologous recombination method was used for CirA-inactivation in Escherichia coli strain CE129, followed by PCR and gene sequencing confirmation. And assays of growth characteristics, biochemical characteristics, antimicrobial susceptibility and biofilm formation ability were performed for assessing the CirA deprivation caused changes. The data demonstrated that, the mutant showed no difference in growth characteristics and biochemical characteristics with wild type strains. But compared to the parent strain, the CirA-deficient mutant displayed sharply impired antibiotic tolerance and biofilm formation. In this study, we successfully generated the cirA knockout mutant of E.coli, and deletion of cirA gene caused increased antibiotic suspectivity.
