Determination and analysis of rDNA-ITS genes from Lasiohelea taiwana

Abstract:The ITS1 and ITS2 genes sequences of Lasiohelea taiwana were amplified and determined to investigate the feasibility of rDNA-ITS in the molecular identification of blood-sucking midges. PCR was employed in the present study and phylogenetic tree was analyzed according to the ITS1 and ITS2 genes sequences. It was showed that the framents of ITS1 and ITS2 are 310 bp and 360 bp, respectively, sharing the greatest homology of 71% and 92% with Forcipomyia townsvillensis. It was demonstrated in the phylogenetic analysis that the molecular identification based on ITS1 and ITS2 gene sequences is consistent with the morphological identification. Therefore, the sequences of rDNA-ITS can be the molecular marker in the molecular identification of biting midges.
