PCR specie identification and phylogenetic position analysis for microorganism in tapeworm from chicken

Abstract:To identify the bacteria and fungi species that were parasitic on Raillietina echinobothrida found in chicken, the present study designed the bacteria 16s rDNA and fungi 18s rDNA universal primer respectively. After extracting the genomic DNA of Raillietina echinobothrida, target genes were amplified by RT-PCR and then sequenced. Additionally, the phylogenetic trees of target gene sequences were constructed by neighbor-joining method and genetic distance were analyzed by MEGA5.1 software. Results showed that the bacteria parasitic on Raillietina echinobothrida belonged to Achromobacter sp. and the identity were 99%. The fungi were identified as Malassezia sp. in the known species and genomic homology was the closest to the Malassezia restricta, reaching to 99%. In conclusion, the present study firstly identified the bacteria and fungi species in Raillietina echinobothrida by PCR method, and the results are expected to provide an effective reference for the prevention and treatment of secondary infection in chicken caused by Raillietina echinobothrida.
