Abstract:To study autophagy induced by Acinetobacter baumannii in Raw264.7 cells, the cells was co-cultured with A. baumannii (ATCC17978) for 180 min, then total RNA and protein was extracted respectively for qPCR and Western blotting analysis. The genes expression of atg3, atg4B, atg5, atg7, atg12, atg16L1, ULK1 and GABARAPL1 in Raw 264.7 cells was determined by qPCR assay, and the exrpession of autophagy related light chain 3 (LC3) and Beclin-1 were determined by Western blotting assay. After co-cultured with A. baumannii for 180 min, the expression of LC3 and Beclin-1 were increased dramatically compared to normal group (F(LC3)=457.2;F(Beclin-1)=43.27,P<0.05). The genes expression of atg3, atg4B, atg5, atg7, atg16L1, ULK1 and GABARAPL1 had no significantly different between infection group and normal group, except atg12. In180 min, the gene expression of atg12 had a dramatical increase compared to normal group (F=208.6,P<0.05). Our results indicated that Raw 264.7 cell authphagy could be induced by A. baumannii, and the possible mechanism is by the ATG12- ATG7- ATG5 signal pathway. However, the exact mechanism would be explored in further study.
