Isolation, identification and genetic evolution analysis of pig-derived gaeta virus from Sichuan,China

Abstract:Getah virus is a zoonotic arbovirus that is widely distributed in Eurasia and the Pacific coast of northern Australia.For the first time, this study was conducted from a pig farm in Sichuan with a mixed infection of pig reproductive and respiratory syndrome blood, RT-RCR test, and detected GETV positive. The samples were aseptically treated, inoculated into BHK21 cells, serially subcultured, and cell cultures were identified by cytopathic, plaque purification, RT-RCR amplification, and whole-genome sequencing.The results showed that we successfully isolated the first pig lidta virus in Sichuan and named it SC201807. The whole gene amplification sequence was sequenced and sequenced. The similarity between 35201 GETV sequences uploaded by SC201807 and Gen Bank was 99.0. %-96.1%, the E2 sequence is highly conserved only in E310 (aspartate → glutamic acid) mutation. Genetic evolution analysis of whole genome nucleotides and E2 amino acid sequences showed that SC201807 strain and Chinese pig source HNJZ-S2 and Japanese mosquito source 15-I-752, 15-I-1105, 14-I-605-C2, 14-I- 605-C1, 12IH26 are closely related and are in the same evolutionary branch. SC201807 strain expands the infection spectrum of pig-derived GETV, and provides materials for related research on pathogenicity, infection and transmission mechanism, environmental microbiology and public health.
