Abstract:The species identification of Nocardia is challenging due to rapidly evolving taxonomy. In the study, MALDI-TOF MS (matrix-assisted laser desorption/ionization mass spectrometry) as an advanced proteomic technology was used for identification of clinical Nocardia isolates. A total of 46 Nocardia isolates were isolated from clinical specimens in the study. Molecular identification of Nocardia spp. was performed using partial 16S rRNA and gyrB gene sequences as the reference standard. Nocardia species identification by VITEK MALDI-TOF MS was explored. As results, forty-three of 46 (93.5%) Nocardia isolates could be identified to species-level, while 45 (97.8%) isolates could be identified to genus-level. The most frequently encountered species, N. cyriacigeorgica and N. farcinica were all identified with 100.0% accuracy (22/22 and 7/7). The results mentioned above indicated that a rapid, accurate, and simplified procedure of Nocardia species identification can be accomplished by VITEK MALDI-TOF MS, and will be used in clinical microbiology laboratories.
