Abstract:In this study, the molecular characteristics of H6N2 subtype avian influenza virus isolated in a poultry market in Guangzhou were analyzed, to provide research data for scientific prevention and control of avian influenza. The whole genome of A/EN/Guangzhou/13565/2018 (H6N2) was sequenced. The molecular characteristics were analyzed with bioinformatics software. The most similar strains of each gene segment of this isolate were strains of the H6N2 avian influenza virus isolated in China. The maximum nucleotide sequence similarity ranged from 96.85% to 98.22%, and that of amino acids ranged from 96.30% to 99.30%. All gene fragments belonged to the Eurasian lineage. The genes HA, PB2, PB1, PA, M and NS belonged to the ST339-like branch, whereas the NP gene belonged to the HN573-like branch, thus suggesting that this isolate recombined from strains from multiple evolutionary sources. The NA gene belonged to the H6N2 subtype avian influenza viruses rather than the H1N2, H3N2, H4N2, H5N2 or H9N2 subtypes. This isolate was an avian-origin virus with low pathogenicity. However, amino acid variations were found at the PB2-89, PB1-473, PB1-F2, PA-409, M1-30/215 and NS1-42/205 sites, which enhance the pathogenicity to mammals. In conclusion, a recombinant H6N2 subtype avian influenza virus, detected in the poultry market in Guangzhou, was found to be an avian-origin virus with low pathogenicity. Because of the potential risk that this strain might infect mammals, continuous monitoring of H6N2 subtype avian influenza virus is needed.
