Comparison of four kits for extracting DNA from histopathological sections containing plerocercoid |

Abstract To screen a rapid, simple, effective and affordable kit for extraction of DNA from the histopathological sections, the DNA was parallel extracted from white/ HE stained slices containing plerocercoid with four kits. By comparing the concentration, purity, extraction rate, time, price and brightness of PCR products, the extraction effect of the four kits was evaluated. Extraction results of white slices showed that the concentration, purity and extraction rate of DNA extracted with kit 1 were the highest, the cost was also the highest but the extration time was the shortest; the extraction effect of kit 2 was similar to that of kit 1, the cost was lower but the extraction time was the shortest; the extraction effect of kit 3 was between kit 2 and kit 4, the cost was the lowest but the extraction time was almost 26 hours; the extraction effect of kit 4 was the lowest and the brightness of PCR products was also darkest, the cost was low but the extraction time was long. Extraction results of HE stained slices: kit 1 was similar to kit 2, both cost were high but extraction time was low, kit 3 was similar to kit 4, both cost was low but extraction time were long, the extraction efficiency of kit 3 and kit 4 was lower than that of kit 1 and kit 2. The four kits can meet the DNA extraction of tissues sections containing plerocercoid, in which kit 2 is more fit for fast extraction.
