Multi-locus sequence typing of Brucella strains in Shaanxi Province |

Abstract Genetic polymorphisms of 77 Brucella strains isolated from Shaanxi Province were analyzed to assess their genetic characteristics and evolutionary relationships. Multiple locus sequence typing (MLST) was used to identify the sequence types (STs), on the basis of seven housekeeping genes, one outer membrane protein gene and one intergene region, in 77 Brucella isolates from Shaanxi Province. A minimum evolutionary tree was constructed to analyze the genetic and evolutionary relationships among the strains in BioNumerics (Version 7.6). The results showed that 74 strains (47 Brucella melitensis biovar 3, ten Brucella melitensis biovar 1, seven Brucella melitensis biovar 2, and ten Brucella melitensis biovar variant) were ST8, two strains (Brucella abortus) were ST2, and one strain (Brucella suis biovar 1) was ST14. The MLST results for the different biological types of Brucella were stable, and Brucella Melitensis biovar 3 (ST8) was found to be the main epidemic strain in Shaanxi Province. MLST genotyping as a technical supplement to traditional biological typing was used to establish a Brucella genotyping database, thus providing scientific guidance for the prevention and control of human brucellosis in Shaanxi Province in the future.
